If i accidentally tore the surface of the agar using the loop during streaking, will the bacteria colonies still able to grow?
Colonies will still grow, yes. However, it is usually best to restart on a fresh plate as the puncture could lead to difficulties in reading/counting colonies or could cause unwanted areas of heavy growth. Don't worry though! Marring the surface is extremely common, especially in beginners. Keep practicing and you'll be able to do it as if it were nothing!

Why did you put the hose inside the autoclave into Erlen?
Thank you for your question. The hose is actually a temperature probe. As the video explains the probe can be placed directly into the media (which is in the Erlenmeyer flask aka “Erlen”). As the directions state: “Place the flask of boiling media into an autoclave to sterilize. If your autoclave is equipped with a temperature probe, the temperature probe can be placed directly into the media and the cycle parameters will be based on the readings of the probe.”

@mel-j The 15 minutes recommendation is part of the general guidelines provided by CLSI for a standardized procedure. It is important to use optimal inoculation suspensions, media, and incubation conditions for the most accurate and repeatable results. Please refer to the CLSI website and guidelines for more information.

not a very good technique by the fellow and the method/procedure used its bad. usually you dont attack directly the sample with stain or water or whatever you pour on it. heat its not bad at all, just not direclty for more than 5 sec, like in flames works good enough... air dry could contaminate your sample and/or could have alive bacteria (wich isnt bad at all either but it depends on what you want, generaly dont want alive bacteria so you dont contaminate more tools in your lab).
